A soft technology to improve survival and reproductive performance of Litopenaeus stylirostris by counterbalancing physiological disturbances associated with handling stress ArchiMer

The consequences of handling stress (fishing, transfer, eyestalk ablation) on shrimp broodstock are poorly documented. The weakness of farmed shrimp, Litopenaeus stylirostris, during winter is a major problem in New Caledonia, because of seasonal climate (tropical-sub-temperate). The transfer of broodstock in winter from earthen outdoor ponds to indoor maturation tanks in the hatchery (T=20 degrees C, Salinity=35 parts per thousand, fed shrimp) usually leads, after 48 h, to high mortality (up to 70%). Eyestalk ablation to induce ovarian maturation in females leads to further mortality. The New-Caledonian shrimp industry is based on the controlled reproduction of the shrimp Litopenaeus stylirostris, a species which was introduced in the 80s. The major difficulty to which the industry has been faced for 10 years is the occurrence of the "syndrome 93", which corresponds to mortality phases when the temperature falls down in April-May-June. This mortality is associated to the pathogenic bacteria Vibrio penaecida and is expressed at different levels which are variable from year ta year and from pond to pond. No resistance to this pathology has been developed spontaneously. Selective breeding of shrimp for viral resistance is established for species like L. vannamei and L. stylirostris. However, selection for bacterial infection resistance is not documented. In New Caledonia, "Syndrome 93" is a vibriosis that affects cultured shrimps during the cool season with survival from 15 to 30%. Current practises for breeders rearing at low densities do not improve the resistance of the offsprings mainly because selection pressures for "Syndrome 93" are low. Even after the fifth generation, survival improvement is not significant.

Biosécurité en écloserie : le rinçage des Artemia ArchiMer

Seawater, algae, micro-diets and Artemia are the main inputs in shrimp larval rearing. Each of them are potential carriers of pathogenic bacteria in the rearing media and weaken the animal in its early life stages. Daily monitoring of rearing water and larvae showed a bacterial spike after the first live feed meal. Vibrio settlement can be enhancedby giving Artemia with their incubation water where vibrio concentration can reach up to 105 CFU per milliliter. This study showed that simple measures such as rinsing and supplying Artemia with freshwater allowed a significant drop in their bacterial load.

New Artemia strain from Russia are now available on the market. They have been evaluated by comparison with Artemia from Great Salt Lake which are routinely used at the LAC hatchery,. Five criteria were compared and all of them were in favour of the American strain with 191 833 nauplii per gram while the Russian ones only gave 102 667 nii per gram. Another important criteria when using Artemia was hatching time which turned out to be longer for the Russian.

Since 1993, the New Caledonian shrimp industry has been faced with cold season mortality outbreaks of multifactorial origin. This pathology called “Syndrome 93” affects as well animals in grow-out ponds as broodstock after their transfer in maturation room, with up to 35% of mortality in 48 hours. From literature data and physiological studies on Litopenaeus stylirostris, several combinations of three abiotic parameters (temperature, salinity and feeding) have been tested during the breeders stocking period. This research allowed us to lower stress factors impact and to propose a transfer protocol which leads to an optimal physiological comfort of the animal. Two populations of the Latin American shrimp Penaeus (Litopenaeus)stylirostris domesticated in Hawaii and in New Caledonia were previously shown to be genetically differentiated and proven highly inbred. New Caledonian shrimp farming feasibility studies started in the mid-seventies. Today ca. 2000 tons of Litopenaeus stylirostris, introduced in 1978, are semi-intensively produced each year. Since fifteen years, production has been impacted by two seasonal vibriosis, the “Summer” syndrome caused by Vibrio nigripulchritudo in relation to pond bottom and water column quality, and the “Winter Syndrome” caused by Vibrio penaeicida in relation to drastic temperature drops . The first aim of this study was to evaluate on a technical basis the rearing of shrimps in floating cages set up in earthen ponds. Shrimps reared in and outside the cages presented comparable growths and survival rates. Then it was concluded that shrimp culture in floating cages did not present zootechnical biases comparing to pond culture. The second objective of this study was to assess the present method as a statistical point of view. We examined the potential for producing the large numbers of sandfish (Holothuria scabra) needed for restocking programmes by co-culturing juveniles with the shrimp Litopenaeus stylirostris in earthen ponds. Our experiments in hapas within shrimp ponds were designed to detect any deleterious effects of sandfish on shrimp, and vice versa. These experiments showed that a high stocking density of juvenile sandfish had no significant effects on growth and survival of shrimp. However, survival and growth of sandfish reared with shrimp for 3 weeks were significantly lower than for sandfish reared alone. The implication of branchial organs in ionic regulation was first demonstrated in juveniles by immunolocalization of three ion transporting proteins ( namely NKA, co-transporter NKCC1 and CFTR ) and ionocyte detection was done by means of electron microscopy. Live preys in shrimp culture: nutritional and sanitary considerations on the use of Artemia in New Caledonia ArchiMer

In aquaculture, Artemia nauplii are commonly used as live prey; and represent 40 % of the total aquaculture feed demands in the early stages. However, the production of Brine shrimp is very unstable from one year to another and in the late 90’s, only 20 % of the world aquaculture need was covered. An insufficient offer leads to higher prices of the Artemia cysts and consequently, the price of shrimp juveniles from hatchery. In New Caledonia, marine aquaculture is mainly based on the blue shrimp Litopenaeus stylirostris. From Zoea 3 to 10 days old post-larvae (P10), the shrimp are mainly fed with live preys. The role of the main ion transporting enzyme Na +/K + − ATPase in osmoregulation processes was investigated in Litopenaeus stylirostris. The development and localization of the osmoregulation sites were studied during ontogenesis by immunodetection of Na+ K+-ATPase using monoclonal antibodies and transmission electron microscopy (TEM). Osmoregulation sites were identified as the pleurae and branchiostegites in the zoeae and mysis stages. In the subsequent post-metamorphic stages the osmoregulatory function was mainly located in the epipodites and branchiostegites and osmotic regulation was later detected in the gills. Probiotic P-acidilactici application in shrimp Litopenaeus stylirostris culture subject to vibriosis in New Caledonia ArchiMer

We studied the effects of a lactic acid bacterium, Pediococcus acidilactici (strain MA 18/5M, CNCM), as a dietary probiotic on growth performance and some nutritional and microbiological aspects of the shrimp Litopenaeus stylirostris. Experimental shrimp culture was carried out over 10 weeks, using floating cages of 14 m(2) each set in earthen ponds, in a New Caledonia farm affected by "Summer syndrome", a septicaemic vibriosis caused by Vibrio nigripulchritudo.
